Stimulate the THP-1 into monocyte - (Oct/31/2005 )

I want to harvest immature dendritic cell by adding cytokine like GM-CSF and IL-4 into monocyte, and now I am trying to do is to stimulate the THP-1 into monocyte like cells. What reagent and concentrate should I add to stimulate THP-1 ? Is there any precaution I should take before I start my experiment?
Any advice will be very useful

THP is human monocyte cell line in itself. If you wish to convert it into macrophage then you will have to try 20nM Phorbol myristic acetic acid. for 24 hours only.
