promoter complex - (Oct/26/2005 )

Say I have a sequence of a promoter, I want to purify protein complex
associated on it. Is there any successful example?
Should I do some crosslink?

Most people do a ChIP assay (see links on this website). Basically proteins are cross-linked to sheered DNA and then immunoprecipitated with antibody to the protein. If your protmoter sequence is enriched by the immunoprecipitation (measured by PCR), then it is assumed that your protein is associated with the promoter.
