conservation of antibodies! - after purification! (Oct/20/2005 )
hi everyone,
I purified antibodies (from rabbit) and I have some trouble with the storage! what kind of storage should I use?
- 4°C
- -20°C
- -20°C with glycerol
- -80°C
I tried -20 and 4 °C they work fine for IF but IB is not that good!

-if you need your antibodies in the next few days, store them with no additive at 4C to avoid a cycle of freezing/defreezin which could damage them...
-storage of concentrated antibodies is ok at -20C
-i have never tried storing lyophilized antibodies, but i think it will be good for -80C
-finally, i generally use hypertonic buffer to store my mAb at 4C for long time:
NaCl 0.3M
Tris 20mM pH7.0
Tween 80 0.02% (max 0.1%)
please make this buffer in sterile condition and without any other protein
After dialysis, mAb are stable for more than 1 year at 4C (do not freeze!)

A modest Tris buffer will do if you store them at -80. Also, although I've never tried, some people add a bit of BSA to help stabilize the antibodies...
We usually store them in PBS at -20 and they are just fine for everything.
For a really long term storage, I would suggest to store them with glycerol [1:1].

What I use to do is to keep my Ab in PBS, when it's for biochimestry or histoschemistry :
to avoid freezing/thawing cycles, I add glycerol 50%.
to prevent adsorption of the side of the tube, I add BSA 0.1%.
to prevent bacterial growth, I add azide 0.05% (not with secondary-HRP because Azide inhibits HRP activity).
In all case I prepare aliquots of 50ul that I store at -20°C.
I've never tried the dehydratation, but I could help for long term storage.
