PCR after Sonication... - (Oct/04/2005 )
I didnt really change anything. It still workes eventhough the cells arent lysed b4 I sonicate. I just get more of a smear compaired to no crosslinking (I would post pics but we still have an old camera to take pics of the gels).
I kept the cells in the lysis buffer for 8h, still no lysis. But the untreated cells (no formaldehyde) lyse instantly (viscous solution, and seen under the microscope).
But my input control works now, and im going to move on to see if I can get the rest of the procedure to work.

how did you work this out? When we lyse our fixed cells we just look for the cloudiness of the suspension from the cells to dissappear which means they have lysed.
