hi,
i would like to know how to conduct a recovery test?
should i spiked a know standard to sample or just sample blank?
for HPLC, can i just compare with peak area?without knowing the concentration of standard that i spiked?

For a recovery study you MUST use authentic matrix (eg. If you intend to use your assay to measure an analyte in serum your recovery studies should be done by spiking serum.
The recovery study should be undertaken by spiking matrix with both a low and a high concentration of analyte. The concentations should be chosen to be around the lowest and highest concentrations you will determine in your assay.
