sox-2 staining - (Jul/16/2009 )

Hi,

I am trying to staing mouse brain sections for sox-2 (frozen, FIXED WITH PFA 4%).
The problem is that I get a lot of positive stainig - not only in the DG and SVZ, but also all over the brain.
It doesnt look like backgroung. Did anyone tryied to use sox-2 antibody ? do you expect to see so many positive cells?

I have never tried Sox2 on the brain. However, it seems that you are not alone (click here).

The staining should be very nuclear and it should only be in areas of neural stem cells (so it should not be everywhere). I an antibody from R and D Systems, Cat # MAB2018 and have never had problems. I have seen Sox2 staining everywhere with other antibodies. GL
