Na2CO3 addition instead of NaHCO3 to RPMI/PS - (May/04/2009 )

Dear all,

a week ago, I accidentally added 2 g of Na2CO3 to my PRMI/Pennicilin Streptomycin stock instead of adding 2 g of NaHCO3. I wonder if anyone had this mistake done before and what happen to your cells? I am maintaining LNCaP and DU145. Morphologically, they look 'normal' as normal as cancer cells can be, but I am worried whether they will have mutation etc that will cause problems in subsequent experiements.

The purpose of NaHCO3 is generally just a pH buffer, and since Na2CO3 is also a salt of an acid and a base, it should also work. That said, I would try to get them in correctly formulated medium as soon as possible, since the equilibria are different. This probably won't affect the cells, but it may have implications for any other manipulations you do, like treatments with drugs.
