what is the activity of TAQ at 60 centigrade? - (Apr/20/2009 )
hi there,
I want to decrease my extension temperature to 60 centigrade.how many bps per second does TAQ do extension?
thank you in addvance.

beckman-coulter recommends an extension temperature of 60C for 4 minutes for cycle sequencing with the genomelab dtcs kit (they use taq). this can generate up to about 900-1000 bases (not necessarily all readable).
this is not to say that taq doesn't work faster (in fact, i'm sure that it does work faster) but this works out to ~250 bases/minute or ~4 bases/second.

THANK YOU VERY MUCH.
My target sequence has high GC% in middle and high AT% in the terminal.
