L.Lactis culture - M17 media (Apr/08/2009 )
hi,
i am planning to grow lactococcus lactis in M17 media.but i went through two protocals one says,we need to grow it in aerobic condition and other artical says it should be grown in anerobic condition...so i am confused..can somebody help me....
Lactococcus lactis is facultative anaerobe. In my lab, if we are going to isolate L. lactis, we normally do it in anaerobic condition. After few subcultures, it can be grown on M17 agar without anaerobic condition and M17 broth as stand culture.

Hi, I have just started my work with Lactococcus lactis, i have 2 strain of it (NZ9000) and (NZ9000 mutant one), i have recived as a stab culture and i have try to revive the culture in M17 broth supplement with 0.5 % glucose and incubate it at 30 degree without shaking but i am not able to find the growth, can you suggest how i can restore my L.lactis culture

Well,its quite wierd that it is;t growing in that condition..i have been working on diff strains of L.Lactis from one year...i have't faced such problem.....u are doing the right thing...why dont u try it with aerobic condition...try culturing it with Sucrose instead of glucose..Practically it should grow in glucose...i used DIFCO M17....autoclaved it and then after cooling i added glucose and then cultured it...I dont know why it is;t growing in urs....check the stab culture again,whether the cells are liable or not...

Is it posible that my stab cluture is too old? or may be i face problem because i used M17 broth from Himedia
I guess the GM 17 from himedia is not a problem..i assume there is a problem with stab culture...try getting new one..it may work out
