Virus storage - (Mar/19/2009 )

Anyone know how best to store viruses long term?

-microgirl-

At -80C or in liquid nitrogen

Do you put it in glycerol like bacterial cultures?

-microgirl-

no glycerol.

Just have them in what ever buffer they are in and freeze it in small aliquots. so you dont have to refreeze many times. But this also depends on the virus.

Depends on the culture media.. pH etc...

What buffer do you recommend? Is PBS ok?

a recommended buffer is SM buffer :
per liter:
NaCl = 5.8g
MgSO8 X 7 H2O = 2g
1M Tris-HCl (pH 7.5) = 50ml
2% (w/v) gelatin solution = 5ml
H2O to 1 liter

sterilize by autoclaving and than dispense into aliquots. Store at RT.

We keep in PBS or TEN buffer and store in -80.
