the product length should be 306bp - the product length should be 306bp (Feb/16/2009 )
hi admin and chatter,
i just run my new primer. the product length should be 306bp and product region 1 -306. but in gel electrophoresis the the product length show ~150bp. why possible happen to my primer..
if u have any idea or got same problem with me, pls share with me
thank you

it could be primer dimer or non specific amplification.
Might want to double check the primer binding site with the template to double confirm the product size.
but seriously 150 bp might just be some form of dimers.

What are the pcr conditions??? and Tm of primers??

i belive that is not primer- dimer if not found under negative control, this size is long on primer- dimer and also if this size of amplicon from plant must less if used with micro organism but not sure

could be some overlapping sequence in ur template which share homology high enough to serve as a templae to then generate the 300+ bp product?
