In vivo transfection of plasmid DNA, how? - intramuscular injection DNA (Jan/26/2009 )

Hallo, can anyone advice me, if it is possible to transfect mose directly by intramuscular injection of DNA plasmid with my structural gene mixed with PBS and transfection agens(lipofect amine). How much DNA PBS and lipofectamine should I use, to which muscle inject an in what interwals. I want to induce antibody and T cell response perhaps. I have no electroporation equipment

Your cloning vector should be a suitable DNA vaccine vector- if that is the case, you can inject it without the need for transfection agents and still elicit an immune response as long as u use the proper adjuvants.

There are a lot publications on this subject. Do a search first. Direct i.m. injection of naked DNA (100 ug/100 ul) can lead to some detectable levels of expression. When combined with electroporation, the expression is greatly elevated.
