RT-PCR - can you keep the PCR plate in the fridge? - Please help! (Jan/26/2009 )

Hey guys,

So glad the forum is back up running! I was just wondering if anyone has any experience of leaving a PCR plate in the fridge prior to running the PCR? Would my plate be ok with the samples and primers in it for about 4 hours before I run it on the pcr machine? And then should I wait for it to return to room temp before running it? Any help would be much appreciated! Thanks!

i haven't done it but there should be no problem holding the plate at 4C until you are ready for it. that's what you do when you are finished with the reaction (4C hold).

you shouldn't need to prewarm the plate to room temperature, the cycler will take care of all temperature changes.
