How to culture PC3 cell line - (Jan/26/2009 )

Hi, I work at Warsaw University in Molecular Biology Department. I just started working with PC3 cell line and I have some questions to somebody who works with this line. What kind of medium do you use? In cell line data shet I found Coon's mosified Ham's F12 medium, but in many publication I foudn RPMI 1640. How often You change the medium? And How often do you passage the cells?

PC-3 cells are pretty easy to culture. We use RPMI-1640 and change medium every 3-4 days.

Is it normal, that colour of the medium changes so much?

And one more question about sub-culturing: how do you subc-ulture these cells? From 70-80% confluent cells 1:5 or rather 1:10?

If the media is changing colour so much you should probably consider performing a media change about half way between passages.
I've done 1:5 splits before.
