cell free DNA (cfDNA) - (Apr/17/2020 )

Hi,

 

Can anyone suggest a good elution buffer for cfDNA. I feel they are more easily degraded (-20deg celsius). We currently elute in deionised water.

 

Qiagen kits elute in AVE buffer, are they better?

Never worked with cfDNA, but with deionised water you have to be sure to have a fitting pH value, otherwise hydrolysis might occur. Buffers with EDTA have also the advantage to chelate  Mg2+ which are needed for enzyme activity (e.g. nucleases).

So an TE-buffer, pH 7 might be appropriate.

Anyway if you elute from a column I'd use the recommended buffer of the company or a clone of it.
