How to calculate cell viability percentage from the absorbance in LDH assay?
I did LDH assay and also get their absorbance at 490 nm and 600 nm by microplane reader. but I don't know how I calculate its viability percentage? kindly help
Sorry about this response being so late, and probably not that helpful (i didn’t do cell culture during my tenure).
wikipedia has a page about viability assays. It contains references which may help you (the article mentions ldh assay)
LDH is rapidly released into the cell culture supernatant when the plasma membrane is damaged, a key feature of cells undergoing apoptosis, necrosis, and other forms of cellular damage. LDH activity can be easily quantified by using the NADH produced during the conversion of lactate to pyruvate to reduce a second compound in a coupled reaction into a product with properties that are easily quantitated.
