activity optimalisation of proteinase K by E. coli - (Oct/25/2018 )

For graduation my bachelors degree in Industrial engineering option biochemistry, I'm currently working on a report about the activity optimalisation of proteinase K (prot k).

The prot k is induced in the plasmid of E. coli bacteria. The goal of is to change the conditions in which the E.coli bacteria are growing, in order to make more active prot k.

My research has let me to believe that the incubation time will affect this. Now is my question, do you know more parameters I can change and test?

If you haven't already read this paper I suggest you start here:

https://febs.onlinelibrary.wiley.com/doi/epdf/10.1111/j.1432-1033.1989.tb14539.x

It describes some of the challenges of expression in E. coli, especially regarding the proteolytic activity being potentially damaging to the host cell. Good luck!
