Can we extract outer membrane protein from stored whole cell protein supernatant ?
membrane proteins usually, and mostly, will pellet with the membrane fragments when centrifuged. you would probably be able to extract them from the pellet, rather than the supernate.
Actually, while reading the procedure of both whole cell protein(WCP) and outer membrane protein(OMP), OMP follows the WCP extraction procedure initially.Later OMP is extracted after some additional steps. Generally the extracted WCP is stored at -20?C. So i think we can extract OMP from WCP after following the additional steps. Is it right ?
Also, WCP is stored AS SUPERNATANT.
if you lysed the tissue with detergent then the supernate should contain membrane proteins.
some proteins don't tolerate freezing. you may have to clarify after thawing before further extraction. but, just follow the procedures that you have.
