In vitro culture saffron - (Aug/25/2017 )

Hi, i make a saffron culture using in vitro technolgy, i used MS medium and i cant have a growth or desinfection, can someone helping me with this?

is the medium autoclaved or sterile filtered?

sucrose concentration in the medium?
