How to calculate non- linear Scatchard plot using adhered cell culture and radio isotope labeled ligand. I am studying the cooperativity in EGFR .
I am asking for the most straight forward/ easy to use method. please guide me to tutorial or any other source that explain this in details.

the scatchard plot should always be linear. if it's not then the ligand binding has not reached equilibrium.
however, it could exhibit multiple linearity if a threshold is reached where the binding protein undergoes a conformational change which causes the binding affinity to change or opens up secondary binding sites.
