Can I use Duran bottle instead of erlenmeyer for fungal fermentation? - (May/11/2016 )

Dear guys,

 

I need your help about my project.

I will do fungal fermentation. Previously I used erlenmeyer 500 ml for the process but unfortunately there are many people in my lab use the same volume of erlen so there are't enough of erllen for my project. I should modify and I think I could use Duran bottle 500 ml.

 

Since the bottle neck is wide, can I used the original cap and cover it with parafilm to avoid contamination? Or I should put cotton plug and close it with alumunium foil?

In terms of keeping it sterile, there is not really a problem.

So it should not be a problem.

 

aeration is a problem compared to erlenmeyers, however for you , doing fermentation, its not a problem.

Are you using 500mL medium? If so, better use a 1L bottle. A loose lid should be fine.

 

Following on pito's comment, if you incubate in a shaker, it won't provide the exact same aeration at the same rpm due to the physical differences. Keep it in mind if you are comparing results as it can be a source of discrepancies.

I will use 250 ml of media and use 500 ml of Duran bottle. 

Pito, so you mean it's okay to use the duran cap (lid) ? Should I seal it with parafilm also or just loosen the lid without parafil sealing?

I am afraid there's contamination because I will do my fermentation for 2 weeks.

A couple of things:

If you're going to use Durans then only use them.   Don't use Erlins for some ferments and not others.   That way you will have consistency of results and won't introduce artefacts in your results.

If you seal the Durans then that'll keep oxygen out but how will you vent CO2 produced?   Given everything is sterile then a cotton bung and aluminium foil will allow the positive pressure of the CO2 created to vent and create a positive pressure environment to help assure that nothing gets in...if everything is sterile to begin with.

well you could use a sterile cotton plug like Astilius already mentions.

You could use the regular caps and just loosely screw them on  (it should not get contaminated, nothing can get it).

But not sure how it is regarding the oxygen, CO2, whether that would get out easily... not sure.

 

Also: keep to 1 type of bottle when doing it!
