What microscope magnification to use to count fibroblasts and lymphoblasts - (Jan/10/2016 )

Hello Everyone!

I have a really burning question. I have just started working with cell culture.

When I count cells on 10x magnification, I have one quantity of cells, when I look at cells on 200x magnification, I see more cells, which is obvious. I would really appreciate the explanation: what magnification should I use to count cells?

I'm assuming here that you are using a hemocytometer to do this?

You should use a 10x objective - 100x magnification, such that one of the large squares bounded by 3 lines takes up most of the field of view. 200x objectives would be rare and far too high a magnification to count cells (you would see 1-5 in a field).
