total protein measurement from insect body - (Jan/04/2016 )

Hello

I want to measure a total protein amount of an insect (beetle or ant)

My question is how do i separate (extract) the proteins and then quantify them:

- what lysis buffer should i use for it if i want to measure it with bradford method?

and what do you think is the best method to measure the total  amount of proteins? bradford, PCA, or something else?

Something like RIPA buffer should work well. Avoid the Bradford, as it is insensitive and reagents go off quickly. I have used the BCA assay with good effect, just be aware that some detergents are not compatible with these sorts of assays, so you need to check the data-sheets for the assays to find out how much you can use.
