We would like to track and detect the replication or S. aureus in human macropha - (Oct/12/2015 )

Hi All,

 

We would like to track and detect the replication or S. aureus in human macrophage. So, the question is which S. aureus genes can be used in the RT-qPCR to relativily quantify this bacteria. 

 

Also, we wonder if we need to isolate the total RNA or the DNA to do this ؟

Hi Mohammed,

 

As you want to track the infection by Staph. aureus on/into the human macrophage, I may suggest to use fluorescence microscopy method, in which you could easily track the bacterial cells through using specific antibodies to Staph. aureus, and also using markers (another Ab) against some of the macrophage cell organelles. You could also transform the bacteria by inserting GFP-expressing vector; and here you could also easily track the infection/bacterium position without fluorescent/Abs staining.
