Solubilizing agarose in soft agar assay for absorbance reading - (Jul/29/2015 )
Does anybody have an in house buffer/protocol for solubilizing the agarose/ agar in a soft agar assay?
I wish to stain my colonies (with haematoxilin or crytal violet) and then solubilize the whole thing and read the absorbance on a plate reader.
I know there are certain kits that inadvertantly solubilze the agarose gel (e.g. CellGlo 2.0 from Promega, but I dont have access to a luminometer). So it can be done.

You could try using an agarase and some gentle heat.
