hey there, i started with my DNA isolation but suppose there is a time constraint then which step is the best for freezing the mixture at -80 c for use on the next day??
can i add sds and then freeze it so that i cn continue with the procedure the next day???
NO. continue on with the prep until you are at the ethanol precipitation step. DNA can be stored in ethanol at -80 C for some time. Don't stop just when you're lysing the cells with SDS. There are lots of active enzymes in the cell debris that might chew up your genomic DNA.
Also, to answer your previous question regarding extraction of gDNA from marine bacteria, I just found this article in Biotechniques March 2015 issue. It's free with no subscription.
I haven't used the protocol myself, but Biotechniques vet the protocols well before putting them in their magazine.
