cell counting - (Feb/24/2015 )

Hello good people

I have cultured HT29 cell line  in T25 flask, trypsinized them with 2 ml trypsin and added 8ml of the culture media making a total of 10ml.I then took 100ul of the cell suspension in to the eppendorf tube then 10ul from the 100ul in to the hemocytometer(attached cells).I  got 1078 cells in 4 grids of 16 squares. How many cells do I have per ml?

The volumes you put into the hemocytometer are irrelevant; so long as you filled it properly the volume under each of the large squares should be 10^-4 ml (which should give you your answer if you think about it a bit), but if you diluted the cells (e.g. if you did trypan blue staining) then that needs to be accounted for.

oh ok.I thought the volume I put in to the hemocytometer really matter. Thanks for that Bob. I am not using trypan blue.
