spore production of lactobacillus - (Jan/21/2015 )
we are doing shake flask studies for production of lactobacillus sporogenes spores (i.e bacillus coagulans). we have pure culture and we use corn steep liquor based media, temperature 37 - 42º C and Initial pH 6.0 and MgSO4 in conc of 0.03% . we tried various manipulations temperature, inoculum size time etc but are unable to see spores even after 48-72 Hrs. we increase temperature to 42º C still no spores.
can anyone recommend what can be done to enhance spore production. we want to take fermentation trials once this shake flask trials are done with
Please, use updated nomenclature. 'Lactobacillus sporogenes' is a non-standing name for Bacillus coagulans since 1939!! Lactobacillus spp. do not form spores.
By the way, magnesium? what you need is manganese.

yes i also use the term bacillus coagulans. we are trying for probiotics and this is well known as lactospore so i gave the old name as its familiar with that name. actually we got the reference from someone of the media we are trying and it mentioned MgSO4. we had tried this same method about a year back and used to get sporulation. however when we are doing it now sporulation is not visible even after 72 yrs with pH decreasing, glucose exhaustion and all so we are very confused.
