Blocking buffer for biotin-labeled EMSA - (Dec/22/2014 )

Hi.

Can someone please advice a good blocking buffer for the detection step of an EMSA using biotin-labeled probes?

Basically, anything with lots of diverse protein except milk. Milk has biotin. I believe there are commercial products made from fish protein, such as Pierce Sea Block.

Thanks. I tried this buffer...

1 L blocking solution / 10X wash solution I
  • 7.3g NaCl,
  • 2.4g Na2HPO4
  • 1g NaH2PO4
  • 50g SDS
  • dH2O to 1 L

 

but the membrane was very dirty when I developed it, so I guess this one is not good.
