How can we expand molecular cloning further? - (Nov/30/2014 )

Hi,

I am wondering what ways i can expand my experiment which was to be able to clone an insert into a plasmid. I have already thought of sequencing, but are there any other ways in which i can expand the procedure?

how could i improve the practical? So how can i take this as the foundation of another project? I have created recombinant plasmids, however some did not work effectively.

The protocols for molecular cloning are pretty well established and have been since the late 1980's, so there shouldn't be any need to "expand" the protocol too much. Often problems are due to user error (e.g. using too much DNA in the ligation) rather than a protocol error.

If you want to move to the next level - have a look for Gibson assembly.
