polymerase with or without proofreading activity used in study nucleotide polymo - (Nov/24/2014 )

Dear all! 

I am studying of polymorphism in a gene of fungi. I cloned gene by phusion polymerase and sequencing. Result showed that gene contain 2 polymorphism. But F12 sequence lack 1 nucleotide C. I'm not sure ploymerase removed C????

There is no way to tell - if you repeat the amplification and sequencing and get the same result then it is very unlikely that it was the result of a polymerase error. Phusion has 3'-5' exonuclease activity so it is pretty good at fixing errors it generates.
