Making a fusion protein - (Aug/20/2014 )
Hi guys
At the moment I have a bit of a concern
I was wondering if any one had a spare mement and could explain to me
why do we need a string of AAAAAA, on our foward and reverse primers
I am cloning a fusion protein and am in the primer design phase
Is there a reason for the A's????|
Woudl appreciate any feedback

Where on the primers do you need this string?
There are usually only three components you need in a primer that will make a product for digestion and subsequent cloning. You need a specific sequence targetedto the sequence you want to amplify, a restriction site, and some bases 5' of the restriction site to give the RE something to bind to. This last bit is possibly what you are referring to.
