Sonication of small PCR amplicon - (Jul/23/2014 )

Hi colleagues,

what happens to an amplicon of, say, 500bp after 1 round or 2 rounds of shearing? Will I get a majority of intact amplicons and an equal distribution of all other fragment sizes or will there be a tailing off with decreasing length? Anyone have any experience with this?

That is very easy to test.
Make your amplicon, split, shear one part 1 round and the other 2 rounds and put on gel. You will see.
