Problem shrna cloning, bad sequencing results - (Jul/09/2014 )

Hello guys,

I'm trying to clone shRNA into pCL20c vector to transduce lentiviruses.

I get enough colonies to select several for miniprep and I check them by PCR before to send to sequence.

But everytime I've got a lot of mutation in my sequences ..

Anybody can help please ?
