thionin extraction - (Jun/04/2014 )

hello everyone

can anybody tell me about thionin extraction from fungal infection induced barley seeds? mol wt 4.5 to 5 kda. Is it possible to give ammonium sulfate cuts?? more importantly, after homogenization, how is it possible to concentrate the protein using methods other than ammonim sulfate cuts ie salting out?

to concentrate without salt (eg ammonium sulfate) you can use an ultrafilter (like this one from millipore, with a 1kDa cutoff).

thanks mdfenko

yes ultrafiltration is a good approach. but I was looking for cheaper methods of concentrating protein from cell lysates. do you think using a spin column is a good idea?

if you mean spin "filtration" then yes. the only possible problem is that the lowest cutoff i can find (at millipore) is 3 kDa. that may be a problem for your protein.

here's the page at millipore for 3 kDa cutoff spin filters.
