Quantification of the immunohistochemical staining slides - (May/31/2014 )

Dear all,

 

I have IHC slides of infected tissue. i wanted to somehow quantify the infection in these tissue.

Any suggestion about software or methode to quantify the IHC stained slides.

There are a number of ways of doing this - you could take area infected per section/slide, so long as you have some way of ensuring that you are always at the same point in the block when you do this (i.e. making sure that one section isn't at the very edge of the specimen, and that you aren't comparing this to one from the middle of a specimen).

 

You can do a scoring system - this involves you either taking pictures of a number of random fields from each specimen/slide or scanning the whole slide, and then scoring them for coverage.  A system like this might work like this: score 1 = minimal infection, score 2= 25%, score 3 = 50%, score 4 = 75% and score 5 =100%.  For these sorts of systems, if you take images it is best if you can get a number of people to do the scoring (randomize the images and don't give identifiers, but do give examples of each score level) and then average the scoring for each image.

Thanks but I am talking about certain online tools or software that might be more precise than our naked eyes. I know for example that people stain for Ki76 protein .

You can try cell profiler (java program), or imageJ for quantitation.
