secretome and zymograms - (May/26/2014 )

Hi everyone

I have a question about the study of secreted enzymes by filamentous fungi: as I have to compare kinetic secretome levels , I should make zymograms. Before them I need a protein gel: which one is better: SDS-Page or a NON denaturat.protein gel?  Do you know any protocol about cellulases? thanks a lot , Veronica

non-denaturing is probably better (depends on what you're trying to show). sds-page will require a renaturing step which may not return full activity.
