Two layer form in probiotic culture after added FOS - (May/08/2014 )

Dear all, 

 

I was prepared probiotic culture by adding fructo-oligosaccharide syrup as prebiotic. However, after added FOS, the culture become two layer, where culture formed on top layer and yellowish layer in the bottom. I was wondering why this will happen? May I know FOS is it unsolvable in the culture?

 fructo-oligosacchariden is Soluble dietary fibre (http://www.ncbi.nlm.nih.gov/pubmed/20119826) . Soluble fibers form gel like structure in water by attracting water molecules. What is color of FOS when you dissolve in water?

If FOS is available as liquid form, higher density could be issue - need to mix properly. If it is in powder form I wonder it has so high affinity for water that it attracts water molecules from culture.

Otherwise probiotic culture carrier medium could be issue.

Which culture medium you are using? You can try MRS Broth. You need to look for FOS concentration too.

 

In this article they have used 2% Lyophilised FOS in MRS Broth.

Thanks for the info! I am using milk to culture my probiotic instead of MRS broth. Cheers.
