How to identify CD8 T cell receptor for a particular antigen - (Apr/07/2014 )

Let's say you have an antigen which you've confirmed, by ELISpot, that it is immunogenic in mice.  How would you go about identifying the T cell receptor(s) that are responding to the antigen?

Step 1: Isolate the Abs to your antigen.

Step 2: Generate new Abs against the variable regions (CDRs) of these Abs.

Hypothetically, the Abs from step 2 should recognize any receptors to which your antigen binds.
