Reference genes in miRNAs - (Mar/22/2014 )

Dear All, 

 

I read many papers about miRNA. In the real time PCR for miRNA detection, I used the miRNA ct values in the ctrl wells as a standard gene. Is this true that in miRNA qPCRs, u do not need a standard gene like in mRNA ?

No, what you described is not correct. You need to have at least one internal control such as sn/snoRNAs. Please read this.

Yes, you will need at least one endogenous control for normalization, but if you use multiple controls, it will be better.

Question, why can't you use GAPDH/B-actin for endogenous controls like you do a lot of time for qrtPCR for miRNA pcr?
