Hello...

I am facing a problem with MCF7 cell lines attachement to slides coated with poly-l-lysine and superfrost slide. I start to smear the solution on glass slide and let it dry afterwds the slide is mounted with coverslip and vectashield mounting medium hard-set and not hardset. but when i try to unmount the coverslip from the slide the cells start to detach from the surface and I end up with no cells on my slide. Any suggestions on why and how can i prevent the cells from detache and remove from the slide?

Why do you mount the coverslips and then remove them?  It is not recommended for cells grown on slides, as they will usually detach or be damaged.
