how to separate organelles from mammalian cells - (Dec/11/2013 )

Hello everyone,

I want to get my target protein which is expressed in the CHO cells. With the help of GFP, I know my protein is localized in the nucleus membrane and ER membrane. Now I want to harvest my cells and separate the nucleus and ER from the cells.  I have no experience on this aspect. And I want to get your advices. Could you tell me some references or some useful websites or protocols to help me deal with this?
