Hydrocortisone vs Water Soluble Hydrocortisone - (Nov/14/2013 )

What is the significance for using water soluble hydrocortisone vs hydrocortisone. I am not looking for an explanation of the molecular difference. Rather, I want to know why I would use one over the other in tissue culture or if they are interchangeable. I have regular hydrocortisone. Howevever, I recently acquired a cell line from another lab and their complete media requires water soluble hydrocortisone. Could I use one in replace of the other?

Solubility of a drug is dependent on the medium it is in. Drugs show different solubilities in water, alcohol, etc.. Drugs are also suspended in different mediums depending upon the drugs mode of entry. If it is via skin, you will need medium that is fat soluble. If it is oral, water soluble can suffice and so on...

I understand that issue with solubility, and I know the solubility is different between the two. I am making tissue culture media and am looking for input from someone who is using the different forms of hydrocortisone to make complete media. I want to know if there is any functional difference between the two or if they both work efficiently.

I am culturing normal keratinocytes isolated from gingival tissues. My Greens media recipe from my supervisor consist of water-soluble hydrocortisone. She specifically stated WATER SOLUBLE hydrocortisone. However, I bought the ethanol-soluble hydrocortisone and have been using it in my culture media over a year with inconsistent growth of cell (using explant technique), now I trouble shooting and I suspect it is because of ethanol( used to dissolve hydrocortisone) in my media causing micelles and difficult for my cell to migrate out from the explant and attach to the flask. I ordered the water soluble hydrocortisone last week and the shipment is delayed. My cells arrived in the lab this week so I wanted to start growing them with regular hydrocortisone which I do have. But according to roxx_gurl's experience, I think it is best I wait until the water soluble hydrocortisone arrives.
