Permeability assay protocol - (Oct/22/2013 )

We are planning on running a permeability assay on primary cultured mesothelial cells.  We have never done this assay before. I've checked the literature and have found several publications on determining permeability in endothelial cells. We will try and follow these protocols.  Before we run this assay on our 'valuable' mesothelial cell culture (they only last one passage), I wanted to ask the community if they have had experience with this assay and what inside tips you might have.

Sorry for the delayed reply……the protocol that I followed can be found in this reference:     Martins-Green, Petreaca and Yao; Methods in Enzymology  443:137, 2008.   Good luck!!
