Extra nucleotides in sequencing... - (Oct/01/2013 )

I am trying to sequencing an empty vector. I got following result. Can anyone explain the presence of 8 extra nucleotides in the obtained sequence? Could it be becuase the next 8 nucleotides (black square) are similar to this extra stretch? Or are these really present in my sequence?

It is hard to tell without the electropherogram, but i would suggest that they are likely to be really there. I trust sequencing more than the literature or notes from other researchers.

Hi,

Thanks for reply. Since there was sequence (identical) available from two clones, I did not bother to post electrogram. They look quite fine as below.

Make that "definitely there."
