pH a Buffer - (Sep/11/2013 )
Why is it necessary to only add about half or 3/4 of the diluent for a buffer, pH, and then add the rest? Why can't I add the full amount and then pH? The buffer consists of dry milk and PBS, with a base increasing the pH. Thank you!

because you will go over the volume for which your components were calculated. therefore, your concentrations will be off.
