cell pellet question - (Jul/07/2013 )

Hi-
I was pelleting 2 million cells of a breast cancer line and the pellet appeared huge relative to what I had seen before. The only difference was that this time the volume that the cells initially were in was 10 mls vs. 1 ml from before. Does that make the pellet post centrifugation appear larger? Not sure if I should count again and repellet? Thanks!

If they are in the same type of tube then they should appear about the same size. You did account for the fact that you have 10 ml rather than just 1?

I would definitely count again.
