are they non-specific band? - (May/26/2013 )

I am student and I did a PCR. The expected band size is 181bp. And I found that there is a fade band below my expected band. Are they non-specific bands? or degradation? Thx

The lower bands appear to me to be specific bands, they could be an isoform of the gene. What kind of PCR (gDNA or cDNA) are you doing?

It is gDNA, how could I avoid it?

So what have you done to optimise the reaction?

It might be worth checking the melting plots to confirm its a specific band; there will likely be a second smaller peak near the main peak.
