SEM Microscopy - (Apr/20/2013 )

Does anyone have a good protocol for using an SEM to view bacterial cells and biofilm. We just got a new SEM (JEOL 6500) and don't have any standard protocols or operating conditions to use. Any input would be much appreciated. We've been using the Poor Man's Cryo technique to mount samples but other than than we haven't been having much luck.

Didn't the company who sold it to you teach you how to do it? Because they usually do. It is not easy to prepare samples to view under SEM, it needs expertise.
