Establishing a primary tissue culture from surgically excised tumor - (Jun/09/2012 )

Hello Everyone,

I am hoping someone can help me find a protocol or design a protocol for establishing a primary culture from a tumor, after it is surgically removed from a dog in a veterinary hospital. I'm trying to figure out the best way to take the tumor, as soon as it is removed from the dog, reduce the likelihood of any biological contamination once it has left the animals body, and finally to a flask by either explant culture or culture by dissagregation.

There are many protocols out there that will work, but I strongly recommend you have a read of Culture of Animal Cells: a laboratory manual by R I Freshney. It has all the techniques you need and excellent advice on this sort of thing, including good ways to keep things sterile and how to disaggregate samples etc.
